The Florentine: Thursday night open late, Strozzina (contemporary section) free from 6-11pm! Great place not just for exhibits but to hang in the courtyard (free wifi), cafe' or bookstore.
Jeff Woo: There's one room in this museum that just takes your breath away: Botticelli's "Birth of Venus" & "Spring." I'd spend 50% of my time in this room :)
Jason Fuller: Often the venue of one off events such as Xmas Market (dec) or chocolate fair (feb) the piazza has a great spot for coffee or a drink outside in the north west corner: Ceasar's bar. Nice view!
Jeff Woo: From this main square, walk past the duomo uphill on Via San Francesco for an amazing sunset view! Or wonder up on Via Panoramico (opposite way)
