Avoid the crowds: Buy a ticket online. Go at 8:45am. Immediately walk up to the Nachtwacht, then the great hall behind it, then the west wing, then down.
Unbelievable collection of Rembrandt and Vermeer. There are also many other stunning objects of art from other dutch artists. Get a Holand Pass and skip the long lines.
A great museum but get there early. At 10 AM on a Tuesday I walked right in. By Noon, there was a line around the building just to get to the queue to purchase tickets.

Forget about getting an audio tour: just download the Rijksmuseum app for Android and iPhone. It not only gives you more in-depth commentary, but also allows you to bookmark things you like.
In the week, if you arrive before 10.30am, then you'll manage to cut a HUGE amount of queue time. We got here at about 10am and had a stress free, great morning there.
Would recommend the media tour where you get audio for paintings. When there's too many people standing in front of it, it can be hard to get to the little sign that explains the painting.Only 5 euro!
The audio guide (5 euro; as of Sep 2015) - it's a smartphone-like visual & audio device, just rent one and share with your company because you could always play it back during your visit.
